Fura-2/AM法测定突触体内游离Ca^(2+)浓度的方法

Estimation of cytosolic free calcium withinsynaptosomes with fluorescene indicator Fura-2

采用新型Ｃａ２＋荧光指示剂Ｆｕｒａ－２，测定冷冻伤性脑水肿时突触体内游离钙的离子浓度。

Intracellular free calcium [Ca^(2+)]i concentration has become a focus of attention for investigators in the field of brain edema. It is the purpose of the present study that the changes of the concentration of the free calcium within cortical neurons were investigated on the model of brain edema induced by freezing injury. [Ca^(2+)]i was determined in calcium fluorescent indicator Fura-2/AM loaded neuronal synaptosomes by using monowave length spectrofluorophotometer. The method for preparing synaptosomes, and the method and mechanism of concentration of cytosolic free calcium using fluorescentindicator Fura-2/AM were studied.
