人IL-5片段的化学合成及单克隆抗体的制备

摘要 目的:利用人工合成的hIL-5多肽片段制备抗IL-5单克隆抗体.方法:从初始原料起,采用固相连接肽等方法,合成了人IL-5羧基端19肽,并耦联到载体蛋白BSA上,以此作为抗原免疫小鼠,采用细胞融合法制备单克隆抗体.结果:成功地合成了hIL-5多肽,获得了七株单抗杂交瘤细胞系,对其中ZH10mAb性质及亚类等特性进行了鉴定.结论:可利用人工合成的多肽作为IL-5的替代物制备其单克隆抗体. Aim: To prepare anti-interleukin-5 (IL-5 ) mono-clonal antibody (mAb) by the synthetic 19 peptide of C-ter-minus of human IL-5 (19p). Methods: The 19P was synthe-sized from protected amino acids by phase Boc-Merrifieldpeptide synthesis method and conjugated with Bovine SerumAlbum (BSA). The BALB/c mouse is immunized with theabove conjugate. The hybridomas were prepared by cell fusion and screened by Enzyme Linked Immunosorbent Assay(ELISA). Results: The seven hybridomas of anti IL-5 werescreened. The classification and subclassification of McAbby 2H10 secreted were identified. Conclusion: The synthetic 19 peptide may he tised as antigen for produce the anti-IL-5 McAb.
