摘要
目的:观察5-氟尿嘧啶(5-FU)对大鼠小肠功能的损害.方法:对照组动物(n=6)直接处死,5-FU组动物(n=24)经腹腔注射5-FU(150mg/kg)后,每8只大鼠分别于注射后第1,3,5日处死,检测大鼠小肠粘膜丙二醛含量,谷胱甘肽过氧化物酶,髓过氧化物酶和双糖酶活性,以及小肠对荧光异硫氰酸盐-葡聚糖4000的通透性.结果:5-FU组动物小肠粘膜丙二醇含量/μmol·g-1在注射后第1,3日(0.40±0.09,0.47±0.14)显著高于对照组(0.27±0.06,P<0.05);谷胱甘肽过氧化物酶活性/nmol·s-1·g-1在注射后第1日(109.0±70.2)显著低于对照组(397.9±148.2,P<0.01);髓过氧化物酶活性/nmol·s-1·g-1在注射后第1,3,5日(36.3±10.3,42.7±12.5,53.3±16.2)均显著高于对照组(7.8±3.7P〈O.01);乳糖酶活性/μmol·s-1·g-1在注射后第5日(87.5±20.5),蔗糖酶和麦芽糖酶活性/μmol·s-1·g-1在注射后第3,5日(197.5±45.2,285.4±89.0;852.5±127.5,920.5±360.1)均显著低于对照组(17.0±62.5,438.6±163.7,1472.8±255.4,P〈0.01);血浆中荧光异硫氰酸盐葡聚糖4000的浓度/mg·L-1在注射后第1,3,5日(7.86±1.22,6.98±1.01?
Aim: To investigate the damage of the small intestinal fu in rats induced by 5-fluorouracil (5-FU). Methods: Control group animals (n= 6) were directly sacrificed.5-FU group animals (n=24) were given a single intraperitoneal injection of 5-FU (150 mg/kg), every 8 rats weresacrificed on day 1, 3 and 5 after injection respectively forthe measurement of malondialdehyde content, glutathioneperoxidase activity, myeloperoxidase activity, disacchari-dase activity in intestinal mucosa and intestinal permeabilityto fluorescein isothiocyanate dextran 4000. Con-clusion: 5-FU may induce damage of the small intestinalfunction.
