血小板源生长因子和脑源性神经营养因子促进NG2蛋白聚糖阳性神经祖细胞向神经元分化

摘要 目的探讨血小板源生长因子(PDGF)和脑源性神经营养因子(BDNF)对NG2蛋白聚糖阳性神经祖细胞(NG2细胞)分化的作用。方法根据文献方法从成年大鼠海马原代培养NG2细胞,7d后添加10~40ng/mlPDGF和/或BDNF继续培养7d,以免疫荧光双重染色法鉴定细胞性质。结果10ng/mlPDGF和BDNF联合处置明显增加Map2a/b、GAD67和GABA表达水平。结论PDGF和BDNF联合处置促进NG2细胞向神经元(特别是GABA能抑制性中间神经元)分化。 Objective To investigate the effects of platelet-derived growth factor (PDGF) and brain-derived neurotrophic factor (BDNF) on the differentiation of the NG2 proteoglycan-positive neural progenitor cells (NG2 cells). Methods NG2 cell primary culture was done from adult rat hippocampus according to the reference. On the 7d, 10-40ng/ml PDGF and/or BDNF were added to the medium and cultured for another 7 days. The characteristics of the cells were identified by double immunofluorescence stain. Results Combined treatment of 10ng/ml PDGF and BDNF increased the expression level of Map2a/b, GAD67 and GABA significantly. Conclusion Combined treatment of PDGF and BDNF promotes the differentiation of NG2 ceils to neurons ( particular GABAergic inhibitory interneurons ).
