Objective To observe the tissue morphology of guinea-pig parotid glands after conventional fractionated radiation delivered to head-neck region. Methods Sixty-seven male guinea pigs were randomly divided into group A~G. Fractionated gamma-rays delivered to head-neck region was applied. According to different radiation dose and observation time, group A~G was set up at total dose 0,10,20,30,40,50Gy and 60 days after 50Gy respectively. The body weight, gland weight, histopathologic change, remaining gland areas and inflammatory cell counts of parotid gland were compared respectively. Results The body weight of guinea pigs increased gradually after transient weight loss in the initial stage of radiation. The gland weight of group G significantly increased compared with other groups (P〈0.05),while there was no difference among group A~F (F=0.997,P=0.43). The average parotid gland areas presented a decreasing trend with the irradiation dosage upgrade. For varying degrees of gland structural changes and inflammatory cell infiltration occurred after radiation. The inflammatory cell counts of group F and group G were significant higher than those of the other groups (P〈0.05),while there was no difference between them (t=0.17,P=0.87).Conclusion There is a positive correlation between irradiation dose and the level of early irradiation damage on parotid gland, and inflammatory reaction induced by irradiation maybe participate in it.
