摘要
目的探讨自拟通咽止呃方对局灶性脑梗死大鼠脑组织损伤的影响及其可能的机制。方法自拟通咽止呃方预防性给药后,采用光化学法建立大鼠局灶性脑梗死模型,观察各组大鼠脑梗死体积、激光多普勒检测梗死中心脑血流量、比色法检测脑组织中SOD、MDA含量。结果模型组大鼠出现明显的神经行为症状,梗死灶明显,大脑皮质脑血流量明显减少,脑内SOD含量显著降低,MDA含量明显升高,与假手术组相比,均有显著性差异(P<0.01~0.05)。自拟通咽止呃方能明显改善模型大鼠的神经行为症状,增加脑血流量,增加脑内SOD含量,减少MDA生成,与模型组相比均具有显著性差异(P<0.01~0.05)。结论自拟通咽止呃方对局灶性脑梗死大鼠脑组织损伤具有保护作用,可能与改善缺血脑组织的血流供应和减轻自由基过氧化损伤等机制有关。
Objective To study the effects and possible mechanisms of Zi ni tong yan zhi er prescription on brain of rats with focal cerebral infarction. Methods To establish the Wistar rats with focal cerebral infarction model by an actinic chemistry way after prophylactic dosage. To observe the volume of cerebral infarction of rat in each group, cerebral blood flow in the middle of infarction was detected though laser-Doppler, meanwhile, the contents of SOD, MDA ,NO in rat brain was determined through colorimetric method. Results The symptoms like visible neuro-behavial symptoms, obviously infarction,cerebral blood flow decreased and the brain tissue SOD content decreased significantly and the content of MDA increased greatly were founded in the model rats. All of those changes were significantly different compared with sham operation group (P〈0.01-0.05).Compared with the model group those symptoms were significantly improved after treatment by tong yan zhi er prescription (P〈0.01-0.05).Conclusion The tong yan zhi er prescription can abate the focal cerebral infarction damage to protect the rat brain, improving the cerebral blood flow and reducing oxidative damage maybe mechanisms of its effects.
