苦参碱对人舌癌Tca-8113细胞增殖抑制及机制研究 被引量:5

摘要 目的探讨分化诱导剂苦参碱(Matrine Mat)对人舌癌Tca-8113细胞系增殖抑制作用及其发生的机制。方法体外培养舌癌TCA-8113细胞,用不同浓度的苦参碱对体外培养的舌癌TCA-8113细胞进行干预。分别采用MTT;细胞周期检测;免疫细胞化学染色、间接免疫荧光联合流式细胞仪定量检测等方法,观察其对舌癌TCA-8113细胞的增殖抑制作用并探讨该作用发生的机制。统计学分析采用12.00统计软件包,计量资料用均数±标准差(x±s)表示,t检验做显著性比较,P<0.05为显著性检验水准。结果经Mat处理后的舌癌TCA-8113细胞生长速度减慢,细胞周期明显阻滞;其PCNA表达明显下降。结论 Mat对舌癌TCA-8113细胞的增殖有显著的抑制作用;其增殖抑制作用发生可能是通过对细胞周期阻滞及调控增殖相关基因表达发生。 Objective To investigate the effects and mechanism of matrine on cell proliferating inhibating in human Tca - 8113 cell line. Methods Different concentrations of Matrine were used in the culture of Tca - 8113 cell line. Proliferation of Tca- 8113 cells were observed by ways of MTT, immunocytoehemistry, indirect immunofluoreseene and flow eytometry, quantitative analysis, etc. At the same time, the expression of PCNA protein was determined by qualitative and quantitative analysis. The data were analyzed by Student's t test using SPSS12.00 software package. Results Proliferation of Tca - 8113 cells was inhibited effectively 'after being treated by different concentrations of Matrine. The rate of GI phase cell was increasing with increased concentration of Matrine. The expression of PCNA was decreasing in the experimental groups by 24 h. Quantitative analysis showed that the average fluorescen intensity of PCNA was falling. Conclusion Matrine has significantly effect on proliferation inhibating of Tca - 8113 cell line in vitro. At same time, Matrine could inhibit the TCA - 8113 cell proliferation by down - regulation gen of PCNA expression.
