摘要
STAT3蛋白是一种重要的转录因子,由多种氨基酸组成,其结构中的主要功能部分是SH2结构域和DNA结合区。在多数人类肿瘤中都发现有STAT3蛋白的表达,尤其是激活状态的STAT3蛋白即磷酸化的STAT3(p-STAT3)。p-STAT3在促进肿瘤细胞增殖、抑制肿瘤细胞凋亡以及促进肿瘤组织血管生成方面发挥巨大的作用,并且与肿瘤的临床生物学行为如肿瘤浸润深度、淋巴结转移及肿瘤细胞的耐药性等相关。文中就此进行综述,并讨论了STAT3在肿瘤个体化治疗中的地位。
The STAT3 (signal transducer and activators of transcription 3 ) protein is a transcription factor made up of many amino acids. Its functional fraction contains 2 domains, transactivation and DNA- binding. The STAT3 protein, especially the activated one, i.e. phosphorylation (p-STAT3) , has been detected in diverse human tumors. And p-STAT3 plays an important role in promoting the proliferation of tumor cells, inhibiting their apoptosis and sustaining their angiogenesis. In addition, it is also correlated with such clinicobiological behaviors as the depth of invasion, lymph node metastasis and drug resistance of cancer cells. This review outlines the above aspects as well as the position of STAT3 in individualized treatment of tumors.
