抑肽酶、氨甲环酸对体外纤维蛋白胶支架中软骨细胞增殖的影响 被引量:1

摘要 目的通过在纤维蛋白胶(FG)中引入不同浓度的抑肽酶和氨甲环酸,探索二者对FG支架中软骨细胞繁殖、表型维持、基质分泌的影响。方法构建标准FG支架细胞复合物和不同浓度抑肽酶及氨甲环酸的改良FG支架细胞复合物,进行体外三维立体培养、扩增。观察软骨细胞在FG支架上的形态变化,绘制细胞生长曲线,观察光镜和透射电镜下组织形态学改变并采用免疫组化S-P法检测Ⅱ型胶原。结果标准组及实验1、2、4、5组镜下可见软骨细胞在空泡内分裂增殖;标准组及实验1、5组透射电镜下可见基质的外分泌;标准组及实验1、2、4、5组在1、2周Ⅱ型胶原免疫组化检测可见空泡内软骨细胞周围出现阳性棕色颗粒;标准组及实验1、5组3条细胞生长曲线相近。结论支架中抑肽酶和氨甲环酸的浓度低于10 000 KIU/ml和20 mg/ml时,对软骨细胞繁殖、表型维持、基质分泌无不良影响。
