摘要
目的观察体外血小板线粒体功能的变化,探讨雌激素抗衰老的机制。方法抽取雌性SD大鼠血小板,每份标本分为空白组和苯甲酸雌二醇(EB)组。观察空白组血小板细胞数量、线粒体△ψm、ATP水平在体外随时间的变化情况。EB组用1×10^-6mol/L浓度的苯甲酸雌二醇孵育1h后测定血小板数量、线粒体膜△ψm、ATP水平,与空白组1h各项指标进行比较。结果体外血小板数量、△ψm和ATP含量随时间推移逐渐降低;1hEB组和空白组血小板线粒体△ψm分别为(1.44±0.10)和(1.17±0.10),ATP分别为(26.43±5.61)和(15.88±3.18),EB组较空白组明显升高,差异有统计学意义(P〈0.05)。结论血小板在体外呈现逐渐衰老和凋亡的过程,雌激素能保护其线粒体功能,从而发挥抗衰老的作用。
Objective To study the changes in the function of in vitro platelet mitochondria and investigate the mitochondrial mechanism of the anti-aging property of estrogen. Methods The platelets were extracted from female Spragne-Dawley (SD) rats, and each specimen was assigned into the blank group and estradiol benzoate (EB) group. The in vitro time-dependent changes in platelet count, mitochondrial membrane potential (△ψm) and the ATP level in the blank group were observed. The platelet count,△ψm and the ATP level in the EB group were determined after incubation with 1 × 10^ -6 mol/L EB for one h,and compared with those in the blank group at first h. Results The in vitro platelet count, △ψm and the ATP level were gradually decreased in a time-dependent manner. In the EB group and blank groups, △ψm was ( 1.44 ± 0.10) and ( 1.17 ±0.10), and the ATP levels were ( 26.43± 5.61 ) and ( 15.88± 3.18) at first h, respectively. The ATP level and △ψm in the blank group were obviously increased as compared with the EB group. Conclusion The platelet becomes aged and decayed in vitro. The estrogen can protect the function of the mitochondria, so as to exert its anti-aging effect. The platelet can be regarded as an ideal biological indicator for study on the mechanism of mitochondrial aging.
