长杆观赏甘蓝下胚轴离体再生研究

摘要 以长杆观赏甘蓝(Brassica oleracea var.longata)实生幼苗下胚轴作为外植体,在附加细胞分裂素6-BA的MS培养基上,通过愈伤组织发生途径获得了再生植株,在研究中发现不同6-BA浓度(1、2、4、6 mg/L)对长杆观赏甘蓝不定芽形成有显著影响,其中在6-BA 4 mg/L培养基上的再生能力最强,每个外植体可再生出25.3个不定芽。此外,还对长杆观赏甘蓝离体培养的愈伤组织再分化过程进行了组织学观察。 In vitro plant plantlet regeneration system was established from hypocotyls explant of long-stem ornamental kale (Brassica oleracea var.longata). The results showed that adventitious shoots could be differentiated from explant via callus on MS medium containing 6-BA,and 6-BA concentrations (1,2,4 and 6 mg/L) demonstrated significant affects on plantlet regeneration. It also exhibited that plantlet regeneration was achieved from explants cultured on MS medium supplemented with 4 mg/L 6-BA, e.g. 25.3 adventious shoots per explant. In addition, adventitious shoot initiated from callus was histologically investigated.
