将高浓度大肠杆菌O6的HAM’sF10悬液置入37℃恒温孵化10、30和180min后,分别稀释、过滤和离心制备大肠杆菌O6悬液、滤液和离心上清液,然后将其在体外与10例健康成年生育男性精子一起孵化0、1、2、4和6h,再采用德国Mika计算机辅助的精子分析系统分析大肠杆菌O6悬液及其滤液和离心上清液体对精子前向运动百分率的影响。结果显示:不同孵化时间的大肠杆菌悬液在体外均能引起人精子前向运动百分率显著降低,但其滤液和离心上清液无类似作用。因此,可以认为大肠杆菌O6在体外对精子的作用在于细菌本身,而细菌毒素并无作用。
The solution, filtrate and supernatant of Echerichia coli O 6 were prepared through the dilution with Ham's F 10 solution, the filtration with 0.22μm millpore filter and the centrifugation 15000 g for 10 minutes. Ejaculates were aseptically obtained by masturbation and prepared by swim-up technique to produce a spermatozoa solution of high motility from 10 healthy fertile men. The spermatozoa solution was incubated with the solution, filter and supernatant of Echerichia coli O 6 at 37℃ in vitro. Motion parameters were measured by an automatic analyzing system provided by stromberg-mika(FRG), a type of computer-analyzed multipleexposure photography. Measurements were carried out at 0, 1, 2, 4 and 6 hours in all specimens. The results showed that the solution Echerichia coli O 6 can significantly reduce human sperm progressive motility. However, the filtrate and supernatant of Echerichia coli O 6 can not affect human sperm progressive motility. It is suggested that Echerichia coli O 6 toxin has not inhibitory effect on human sperm progressive motility in vitro.
