七叶皂苷钠对胶质瘤细胞增生和血管内皮生长因子表达的影响

摘要 目的探讨β-七叶皂苷钠对胶质瘤细胞增生及血管内皮生长因子(VEGF)表达的影响。方法将10、20、30、40、50mg/L的β-七叶皂苷钠作用于体外培养的U251胶质瘤细胞并设立空白对照组,四甲基偶氮唑盐(MTr)法检测细胞生长情况,流式细胞术(FCM)分析细胞周期的分布;免疫细胞化学检测增生细胞核抗原(PCNA)和VEGF表达。结果与对照组相比,β-七叶皂苷钠明显抑制U251细胞增生,对细胞有G1期阻滞作用,其抑制率呈时间和剂量效应关系(P〈0.05)。β-七叶皂苷钠作用后U251细胞PCNA、VEGF表达降低(P〈0.05)。结论β-七叶皂苷钠可抑制胶质瘤细胞增生和VEGF表达。 Objective To study the effect of sodium aescinate on vascular endothelial growth factor (VEGF) expression and cell proliferation in glioma cells. Methods In vitro cultured human U251 glioma cells were incubated with 10, 20, 30, 40, 50 mg/L sodium aescinate. Methyl thiazolyl tetrazolium (MTT) assay was used to evaluate cell growth. Cell cycle distribution was evaluated by flow cytometry. Expression of proliferating cell nuclear antigen (PCNA) and VEGF proteins were determined by immunocytochemistry. Results Compared with the control group, sodium aescinate significantly inhibited the growth and proliferation of U251 ceils in time-dependent and dose-dependent manner (P 〈0.05). After treatment with sodium aescinate, the expression of PCNA and VEGF were significantly inhibited (P 〈0.05). Conclusion Sodium aescinate maybe have a therapeutic effect on glioma growth through down-regulating VEGF expression and cell proliferation.
