摘要
本实验目的探讨具有相同碳原子长度的亚麻酸、亚油酸及油酸处理BEL-7402人肝癌细胞后对DNA合成和甲胎蛋白(AFP)分泌抑制的影响。放射免疫法检测肝癌细胞分泌甲胎蛋白(AFP)含量,3H-TdR掺入法测定DNA合成放射活性(CPM)。实验结果表明,三种脂肪酸能抑制肝癌细胞DNA合成,其中亚麻酸、亚油酸抑制作用明显,有显著性差异(P<0.01),油酸作用较弱(P>0.05);40-50μg/ml亚麻酸能明显地抑制肝癌细胞AFP分泌(P<0.05),亚油酸和油酸与对照相比无显著性差异(P>0.05)。结果显示,三种脂肪酸分子中含有数量不同的双键,脂肪酸对肝癌细胞DNA合成和甲胎蛋白分泌的抑制作用存在构效关系,与分子中双键数量有关。本研究扩充了脂肪酸的用途。
The purpose of this study was to investigate the inhibition effect of DNA synthesis and the AFP secretion on BEL-7402 human hepatocarcinoma cells in the presence of linolenic acid,linoleic acid and oleic acid which having the same carbon chains.The AFP concentration was determinated with radioimmunoassay,while the DNA synthesis was studied by the 3H-TdR incorporation(CPM).The results indicated that three free fatty acids could inhibite the DNA synthesis of human hepatocarcinoma cells.The effects of linolenic acid and linoleic acid(P<0.01) were significant,which were stronger than that of oleic acid(P>0.05). 40-50 μg/ml concentration of linolenic acid could inhibite the AFP secretion of hepatocarcinoma cells obviously compare with the blank (P< 0.05).The same concentration of linoleic acid and oleic acid had no difference compare with the blank (P>0.05). The conclusion showed that there was a relationship between the molecular structure and the inhibition effects of three fatty acids containing different numbers of double -bond.
