摘要
目的研究急性肺损伤(ALI)大鼠肺Clara细胞蛋白(CCSP)的变化及相关影响因素。方法将6周龄雄性SD大鼠随机分成4h实验组及对照组、12h实验组及对照组(各8只);实验组腹腔注射(IP)脂多糖(10mg/kg),对照组IP等量生理盐水;于实验第4、12小时观察肺泡灌洗液(BALF)中WBC及分类、肺组织湿/干重比(W/D)、肺组织病理学评分(LIS);检测肺组织γ干扰素(IFN-γ)、白介素8(IL-8)、巨噬细胞炎性蛋白2(MIP-2)含量及BALF中CCSP含量、肺组织CCSP mRNA表达。结果腹腔注射脂多糖4h后,肺部病理改变显著,BALF中CCSP及CCSP mRNA显著减少,肺组织MIP-2增高;12h后,进一步出现肺组织IFN-γ、IL-8显著增高。结论ALI大鼠肺CCSP减少并与急性炎症反应有关。
Objective To study the changes of pulmonary Clara cell secretory protein(CCSP) and associated factors in rats with acute lung injury. Methods Rats were randomly divided into four groups:4-hr-experimental group,4-hr-control group,12-hr-experimental group,and 12-hr-control group(n = 8,each group) . Experimental rats received peritoneally injected lipopolysaccharides(10 mg /kg) and control rats were treated with similar quantity of normal saline(5 ml /kg) . After 4 hrs and 12 hrs the following parameters were evaluated:leucocyte count and class in bronchial alveolar lavage fluid(BALF) ,wet /dry(W /D) lung weight and lung injury score(LIS) ,the concentrations of IFN-γ、 IL-8、 MIP-2 in lung tissues,CCSP content in BALF and CCSP mRNA expression in lung tissue. Results The leucocyte counts and percent of neutrophils in BALF,MIP-2 in the lung tissue,W /D and LIS increased significantly. However,CCSP content in BALF and CCSP mRNA in the lungs decreased in 4-hr-experimental group compared with control groups. But in 12-hr-experimental group,IFN-γ and IL-8 in the lung tissues were also significantly higher in addition to the above changes. Conclusions Pulmonary CCSP decreased in rats with acute lung injury,and it was probably associated with topical acute inflammation.
