摘要
目的研究大鼠自体骨骼肌去神经游离移植成活过程及机制。方法将大鼠骨骼肌去神经2周后游离移植于背部肌肉内,应用组织学HE染色、电镜及酶学方法于不同时期观察移植肌琥珀酸脱氢酶及组织学的改变。结果去神经后组织代谢降低,肌萎缩。移植后2周,外周可见新生血管,中央出现肌凝及肌溶;移植后4周,肌细胞增生,小血管增多,SDH酶示代谢增强;8周后,肌肉再生明显,乙酰胆碱脂酶染色未见再生运动终板;12周后见神经再生。结论肌肉去神经游离移植有利于移植肌肉的有效成活,为临床应用提供了实验依据。
Objective To investigate the muscle survive process and mechanism of free grafting of rats' skeletal muscle after being denervated. Methods Rats' skeletal muscles were free grafted into their back muscle at 2 weeks after denervated, then histological HE staining, electron microscope and enzymology examinations in different periods were used to detect the SDH and histological changes of the grafted muscle. Results The muscles shrunk and their metabolism reduced after being denervated; new vessels existed around the muscles, but coagulate and dissolve the grafted muscle of appeared in the center at 2 weeks after grafting; myocyte proliferation, increase of small vessels and enhancement of SDH appeaved were at 4 weeks after grafting; obvious regeneration of the muscles appeaved at 8 weeks later. The nerve regeneration appeared at 12 weeks. Conclusion The denerration for free muscle grafting is beneficial for the muscle surviving effectively which prviods an experimental basis for clinical application.
