摘要
目的建立安全有效的小鼠胚胎成纤维细胞饲养层体系,用于人胚胎干细胞的培养和研究。方法取ICR小鼠胚胎制备原代成纤维细胞,观察不同剂量的γ射线对成纤维细胞的抑制作用,用流式细胞仪分析MEF细胞周期的改变,用MTT测定细胞的数量。结果在30 Gy剂量的γ射线条件下,能有效地抑制小鼠胚胎成纤维细胞的分裂,且不影响其活力。结论在合适剂量的γ射线照射下,能安全有效的抑制MEF的有丝分裂,为人胚胎干细胞的研究奠定基础。
Objective To establish mouse embryonic fibroblast (MEF) cells as feeder cells in order to culture human embryonic stem cells. Methods The mouse primary embryonic fibroblasts were isolated from mouse fetus of ICR. To investigate the inhibitory action of different doses of γ -irradiation on MEFs. Flow cytometry was used to detect DNA contents of the cell cycle. The proliferation of MEF were observed by measuring cell numbers using MTT assay. Results In the condition of 30Gy, It can inhibit effectively the division of the MEFs and not affect vitality of the MEFs.Conclution In this study, the mitosis of the mouse embryonic fibroblasts were inhibited in effectively and securely in optional dose of γ-irradiation. Which were successfully established for studying of human embryonic stem cells.
