摘要
目的:建立骨筋速康片中士的宁和马钱子碱的含量测定方法。方法:采用高效液相色谱法,Agilent ZorbaxTMS B-C18分析柱(250mm×4.6mm,5μm),流动相为0.01mol·L-1庚烷磺酸钠与0.02mol·L-1磷酸二氢钾的等体积混合溶液(用10%磷酸调节pH值2.8)-乙腈(815:185);流速1.0mL·min-1;检测波长260nm。结果:士的宁和马钱子碱在0.06~0.14mg·mL-1和0.03~0.07mg·mL-1浓度范围内与峰面积呈良好的线性关系。平均加样回收率士的宁为101.09%(RSD=0.66%),马钱子碱为100.19%(RSD=0.68%)。结论:本方法简便、准确、快速,可用于该制剂的质量控制。
Objective: To establish a method for the content determination of brucine and strychnine in gujinsukang tablet by HPLC. Methods: An Agilent ZorbaxTM SB-C18column (250 mm ×4.6mm,5μm) was used for separation. The mobile phase consisted of the equal volume of 0.01 mol·L^-1 sodium heptanesulfonate and 0.02 mol·L^-1 potassium dihydrogen phosphate sodium (adjusted pH = 2.8 by 10% phosphoric acid) -acetonitrile (815 : 185), the flow rate was 1.0 mL · min^-1. The eluate was monitored by detector at wavelength of 260 nm. Results: The linear ranges of bmcine and strychnine were 0.06 -0.14 mg·mL^-1(r2=0.999 6) and 0.03 - 0.07 mg· mL^-1(P = 0.999 8), respectively. The mean recoveries were 101.09% (RSD = 0.66%) and 100.19% (RSD = 0.68%), respectively. Conclusion: The method is simple, accurate and reliable for the quality control of gujinsukang tablet.
