摘要
目的通过改进大鼠心肌缺血/再灌注模型制备方法,提高造模成功率。方法雌雄各半Wist-ar大鼠40只,随机分为伪手术(S)组20只和心肌缺血再灌注(I/R)组20只。整个实验过程在人工机械通气下完成,所有操作尽量不破坏动物组织结构。结果I/R组结扎心肌左冠状动脉前室间支后心电图ST段弓背向上抬高0.4 mV以上,S组手术前后无变化;I/R组造模成功率为90%。结论本方法制备大鼠心肌缺血/再灌注模型简便易行、心脏暴露好、结扎可靠、结果稳定。
Objective To improve the method of establishing in vivo myocardial ischemic-reperfusion rat model and to increase the success rate. Methods Forty (20 male, 20 female) Wistar rats were randomly divided into Sham group (n= 20) and myocardial ischemic-reperfusion group (I/R, n= 20). In I/R group , coronary occlusion for 45 rain was followed by 3 hours reperfusion of left coronary artery. In the period of experiments, rats breathed under the control of artificial respirator. Results The 90 percent of total rats were made myocardial isehemia-reperfusion model successfully. In I/R group., the ST segment elevated more than 0.4 mV. Conclusion The method is simple, convenient and the experiment results are reliable.
