摘要
采用长大杂母猪原代培养的黄体细胞,通过MTT法、DNA Ladder和TUNEL检测法,探讨了0、0.4、2、10、50mg/L棉酚时猪原代培养黄体细胞增殖和凋亡的影响。结果表明,棉酚分别染毒24、48h后,与对照组比较,各浓度组对黄体细胞增殖具有明显的抑制作用(P〈0.05,P〈0.01),黄体细胞的凋亡率达5%~23%;这种细胞毒性作用呈剂量相关性,在一定剂量下,具有时间依赖关系。
This study was designed to determine the effect of gossypol at 0.4,2,10,50 mg/L on proliferation and apoptosis in primary luteal cells of Landrace-Yorkshire Hybrid sows by MTT and TUNEL assay. The results showed that 24 h and 48 h after treatment with gossypol,obvious inhibition effect on luteal cell proliferation was observed in each concentration group (P〈0.05, P〈0.01). The apoptosis percentage of luteal cells induced by gossypol was among 5% to 23 % in each contamination group. Gossypol at high and low dose could inhibit the proliferation of luteal cells and induce their apoptosis. This role was dose-related,and in a certain dosage,a time-dependent manner.
