五加补骨方对模拟失重大鼠股骨无机元素含量的影响 被引量:2

Interventional effect of Wujiabugu Formula on femur contents of inorganic elements in rats after simulated weightlessness*

摘要 目的研究五加补骨方对3周模拟失重大鼠股骨钙及其他元素含量变化的干预作用,初步观察模拟失重情况下,该方对外源钙(牡蛎醋酸水解物)的协同作用。方法雄性W istar大鼠30只,随机分为空白组、模型组、中药组,适应1周,尾吊模拟失重3周。用电感耦合等离子体光谱法测定各组动物股骨钙(Ca)、磷(P)、镁(Mg)、锌(Zn)、锶(Sr)的元素含量。结果与空白组比较,模型组动物股骨Ca、P、Mg、Zn、Sr含量均极显著下降(P<0.001);中药组股骨Ca、P、Mg等宏量元素含量极显著下降(P<0.001),微量元素Zn和Sr含量亦呈显著下降(P<0.01或P<0.05)。与模型组比较,中药组Ca、Mg、Zn、Sr含量均有显著改善(P<0.05或P<0.01)。模型组股骨中Ca、P摩尔比(Ca/P)和Ca、Mg摩尔比(Ca/Mg)都呈下降趋势,中药组Ca/P和Ca/Mg比值均较模型组有提高趋势。结论3周模拟失重可造成模型动物股骨Ca及其他多种元素含量显著下降,五加补骨方能够有效干预这种改变;另外提示单纯补钙不足以对抗模拟失重造成的承重骨骨量丢失,复方药效优于单味牡蛎,并可能促进了机体对牡蛎的利用。 Objective To study the interventional effect of Wujiabugu Formula on the content changes of femur calcium (Ca) and other inorganic elements in rats after simulated weightlessness for 3 weeks, and observe the cooperation between the formula and exogenous calcium (oyster acetate hydrolysate). Method Male Wistar rats (n = 30 ) were randomly divided into the blank group, model group and Wujiabugu Formula group. After adaptation for 1 week the tail-suspended simulated weightlessness was conducted in rats for 3 weeks. The contents of femur Ca, phosphorus (P), Magnesium (Mg), zinc (Zn) and strontium (Sr) were detected by using inductively coupled plasma atomic emission spectrometry (ICP-AES) in all groups. Result The contents of femur Ca, P, Mg, Zn and Sr decreased very significantly in the model group compared with those in the blank group (P 〈0. 001 ). In Wujiabugu Formula group the contents of some maeroelements-Ca, P and Mg, decreased very significantly (P 〈 0. 001 ) , and the contents of some microelcments-Zn and Sr, decreased significantly (P 〈 0.01 or P 〈 0.05 ) but all of them were higher than those in the model group. The contents of femur Ca, Mg, Zn and Sr were improved significantly in Wujiabugu Formula group compared with those in the model group (P 〈0.05 or P〈0.01 ). The molar ratio of femur Ca to P (Ca/P) and molar ratio of femur Ca to Mg (Ca/ Mg) had a downward trend in the model group, and Ca/P and Ca/Mg had an upward trend in Wujiabugu Formula group. Conclusion The simulated weightlessness for 3 weeks reduced significantly the contents of femur Ca and other inorganic elements in animal model, and Wujiabugu Formula can intervene this kind of change. The result indicated that simple calcium supplementation was not enough to antagonize the bone loss of weight-bearing bone induced by simulated weightlessness. The effect of mixture formula is better than that of ovster and mixture formula can improve the biological utilization of oyster.
