摘要
目的观察川芎防风白芷方中主成分防风色原酮Ⅰ葡萄糖苷、防风色原酮Ⅱ葡萄糖苷、防风色原酮Ⅰ、防风色原酮Ⅲ葡萄糖苷、藁本内酯、阿魏酸、欧前胡素及其主成分组合对离体大鼠胸主动脉环舒缩功能的影响。方法根据川芎防风白芷方的饮片配比,结合各主成分在饮片中的平均含量,确定主成分组合样品的组成和配比。采用测定离体血管环张力的实验方法,以氯化钾(KC l)45mmol/L预收缩大鼠胸主动脉环,采用累积加药的方式,观察川芎防风白芷方主成分及其组合对KC l预收缩大鼠胸主动脉环张力的影响。结果以KC l 45 mmol/L预收缩时,川芎防风白芷方中各主成分对大鼠胸主动脉环均有不同程度的舒张作用,藁本内酯、欧前胡素可使KC l引起收缩的大鼠胸主动脉完全舒张,且以欧前胡素的半数有效浓度(EC50)最小、作用最强(P<0.05)。另外,主成分组合样品也可使KC l预收缩的血管环完全舒张,且主成分组合所含各主成分中除藁本内酯外均低于其起效浓度。结论主成分组合样品对KC l引起的大鼠胸主动脉血管环收缩有较强的抑制作用,其作用优于主成分单独应用。
Objective To observe the effects of principal constituents, including prim-o-glucosglcinmifugin, 4'-o-β-gluco-syl-5-o-methtlvisamminol, cimifugin, sec-o-glucosylhamaudol, ligustilide, ferulic acid and imperatorin of Chuanxiongfangfengbaizhi Formula(CF), and their combination on the diastolic and systolic functions of rat isolated thoracic aorta rings. Methods The composition and ratio of combination sample of CF principal constituents were determined integrated the average content of CF principal constituents according to the decoction piece ratio of CF. The precontraction of rat isolated thoracic aorta rings was induced by 45 mmol/L kalium chloride (KCl) by applying the detecting technique of rat isolated artery ring tension. The effects of principal constituents of CF and their combination on isolated artery ring tension were observed by using accumulative increase method. Results After the precontraction by KCI all principal constituents of CF had diastolic effects on rat isolated thoracic aorta rings in varying degrees. E and G induced a complete diastole in systolic artery rings induced by KCl, and the median effective concentration (EC50) of G was the lowest and its effect was the highest ( P 〈 0.05 ). The combination sample of CF principal constituents could induce a complete diastole in systolic artery rings induced by KCl too. The work concentration of CF principal constituents in the principal constituent combination, except E, was lower than the work concentration of one principal constituent used alone. Conclusion The combination sample of CF principal constituents had a strong inhibitory effect on the systole of rat isolated artery rings, which was better than the effect of one principal constituent used alone.
