大黄酸对白细胞介素-6诱导的血管平滑肌细胞增殖的影响 被引量:8

摘要 目的:研究大黄酸对白细胞介素-6(IL-6)刺激的血管平滑肌细胞(VSMCs)增殖效应的影响。方法:(1)采用原代细胞培养方法建立大鼠VSMCs细胞模型,形态学倒置相差显微镜和透射电镜观察的方法进行细胞鉴定;(2)以四甲基偶氮唑盐(MTT)比色法观察不同浓度和不同时间的IL-6对VSMCs增殖能力的影响,以及不同浓度的大黄酸作用12小时对IL-6效应的干预作用。结果:IL-6能显著促进VSMCs增殖,促增殖效应在48h末达峰值且IL-6浓度在0~50.0ng/ml之间呈剂量依赖关系,差异具有统计学意义(P<0.05);而大黄酸可抑制IL-6诱导的VSMCs的增殖效应(P<0.01)。结论:IL-6可刺激VSMCs增殖,而大黄酸能够抑制IL-6的促增殖作用,有望用于干预动脉粥样硬化的发生发展,其机制可能与线粒体依赖的凋亡机制有关。 Objective: To observe the effect of rhein on IL-6-induced proliferation of vascular smooth muscle cells (VSMCs). Methods: By using primary cell culture and MTT colorimetry, the effects of IL-6 and rhein were detected at various concentrations on VSMCs proliferation. Results: IL-6 can significantly promote VSMCs proliferation in a dose-and time-dependent manner, whose maximum effect appears at 50 ng/ml and 72 hours (P〈 0.05). In additiom, rhein suppressed IL-6-induced VSMCs proliferation. (P〈0.05). Conelusion: IL-6 can induce the proliferation of VSMCs, and rhein can inhibit this effect, rhein might be used to treat atherosclerosis, and the mechanism should be involved in mitochondria-dependent apoptosis.
