摘要
以马尾松春梢嫩茎为试验材料,研究了外植体大小及处理方式、培养基类型、激素种类和浓度对马尾松嫩茎愈伤组织诱导和增殖的影响,结果表明:取春季嫩茎为材料,外植体大小以小于2mm的材料随机接种,在改良GD培养基中添加0.30、0.50mg/L的NAA和0.50、2.00mg/L的2,4-D,均有利于马尾松嫩茎愈伤组织的诱导和增殖。2个最适诱导和增殖培养基配方是:改良GD+0.50mg/LNAA+2.00mg/L2,4-D+0.50mg/L6-BA+2.00mg/LKT和改良GD+0.50mg/LNAA+0.50mg/L2,4-D+2.00mg/L6-BA,其中NAA和2,4-D是诱导马尾松嫩茎愈伤组织所必需的植物激素,较高的NAA生长素浓度(O.50mg/L)和适当的2,4-D植物激素浓度(O.50~2.00mg/L)有利于愈伤组织的诱导和增殖。
Effects of the explant size, treatment,medium and hormone on the callus induction and proliferation of the young stem of Pinus massoniana were studied. Results indicated that explants less than 2 mm on the modified GD medium with NAA(0. 30,0. 50 mg/L) and 2,4-D(0. 50,2.00 mg/L) were suitable for callus induction and proliferation. Two better media were modified GD medium with 0.50 mg/L NAA and 2. 00 mg/L 2,4-D and 0. 50 mg/L 6-BA and 2. 00 mg/L KT or modified GD medium with 0. 50 mg/L NAA and 0. 50 mg/L 2,4-D and 2. 00 mg/L 6-BAo NAA and 2,4-D are necessary plant hormone for callus induction. The NAA with higher concentration(0. 50 mg/L) and 2,4-D with appropriate hormones concentration(0. 50 - 2.00 mg/L) are conducive to callus induction and proliferation. These findings provide foundation for further culturing some elite clones of Pinus massoniana.
