流式细胞术检测精索静脉曲张患者精子线粒体膜电位的研究

摘要 目的应用流式细胞术检测精索静脉曲张(VC)患者精子线粒体膜电位并探讨其临床意义。方法将67例精索静脉曲张患者分为VC1组(精索静脉曲张1度,n=26),VC2组(精索静脉曲张2度,n=21)和VC3组(精索静脉曲张3度,n=20),以正常生育男性为正常对照组(n=29)。通过计算机辅助精液分析系统进行精液常规分析。精液标本洗涤处理后用荧光染料罗丹明(Rh223)和碘化丙锭(PI)染色后上流式细胞仪分析,检测精子线粒体膜电位(Rh123^+/PI^-%)。结果VC1,VC2和VC3组精子线粒体膜电位分别为(50.92±17.62)%,(41.34±16.78)%,(26.36±13.61)%,均显著低于正常对照组[(65.12±13.86)%,P〈0.05或P〈0.01]。96例标本中,精子线粒体膜电位与(a+b)级精子百分率呈显著正相关(r=0.647,P=0.000),而与其他参数无显著相关性(P〉0.05)。结论精索静脉曲张可引起精子线粒体膜电位降低,可能是导致男性不育的重要原因之一。 Objective To detect sperm mitochondrial membrane potential (MMP) of varicocele patients by flow cytometry and investigate its clinical significance. Methods Sixty-seven varicocele patients were divided into VC1 group (grade 1,n = 26),VC2 group (grade 2,n=21) and VC3 group (grade 3,n=20). Normal fertile volunteers were selected as control group (n= 29). Conventional semen analysis was performed by computer-assisted semen analysis (CASA). Semen samples were washed and then Rh123/PI staining were carried out to evaluate sperm MMP (Rh123^+/PI^- %) by flow cytometry. Results The sperm MMP of VC1 ,VC2 and VC3 group were(50.92± 17.62)%, (41.34± 16.78)% and (26.36± 13.61 ) % respectively. Lower than that of the control significantly[(65.12± 13.86) % ,P〈0. 05 or P〈0. 01]. There was significant positive correlations between the MMP and the percentage of grade (a+b) sperm(r=0. 647,P=0. 000) ,but no significant positive correlations with other parameters(P〉0.05). Conclusion The decreased MMP occurs in the sperm of men with varicoceles,which might be a risk factors of male infertility.
