摘要
目的应用RNA干扰技术靶向阻断Survivin基因表达,观察其mRNA表达水平及细胞增殖的变化,探讨其对Me3细胞的抑制作用。方法将RNA干扰表达载体Pgenesil—sur通过脂质体介导转染Mc3细胞,采用半定量PCR检测Survivin mRNA表达量;MTT法检测对黏液表皮样癌细胞的增殖抑制作用。结果RNA干扰组Survivin mRNA表达水平(0.61±0.10)比对照组Survivin mRNA表达水平(3.20±0.11)显著减低,差异有统计学意义(P〈0.01),转染Pgenesil-1空载体组Survivin mRNA表达水平(3.18±0.12)与对照组比较,差异无统计学意义(P〉0.0S)。MTT结果表明转染Pgenesil—sur后细胞增殖受到明显抑制,抑制率达81%;转染Pgenesil-1空载体细胞未受到抑制。结论靶向Survivin RNA干扰能阻断肿瘤细胞中Survivin基因mRNA表达,并有效抑制体外培养的Mc3细胞的增殖作用。
Objective Target blocking the expression of Survivin gene by RNA interference skill to approach the depressant. Effect of its mRNA expression level on mucoepidermoid carcinoma cells. Methods Transfected RNA interference expression vector to human mucoepidermoid carcinoma cells mediated by lipidosome ,detected expression of Survivin by semiquantitative PCR. Results Expression of Survivin mRNA was significantly lighter (P〈0. 01) with the interference group (0. 61±0. 10) and the control group(3.20±0. 11). There was no statistically meaningful difference(P〉0. 05) of Survivin mRNA expression between the Pgenesil-1 transfected group(3.18±0.12) and the control group. MTT results showed significant inhibit of cell proliferation after transfection of Pgenesilur,high to 81%. However,non restrain was observated of Pgenesil-1 transfected cells. Conclusion Target Survivin RNA intervention can block the expression of Survivin gene mRNA of tumor cells,and effectively inhibited the proliferation of human mucoepidermoid carcinoma cells vitro cultured.
