摘要
目的:建立HPLC法测定西藏不同产地的铁棒锤、榜嘎中双酯型生物碱的含量方法。方法:色谱柱:Aichrom^(TM)C_(18)(4.6mm×150mm,5μm);柱温:35℃;流动相:甲醇-0.05mol·L^(-1)磷酸二氢钾-醋酸-异丙醇(67:173:4:4);检测波长:230nm;流速:1.0mL·min^(-1)。结果:乌头碱在0.09632~0.4816μg(r=0.9998)范围内呈线性。乌头碱加样回收率为96.3%,RSD为1.87%(n=6)。结论:方法可控,结果稳定,为乌头类药材含量测定提供一个参考方法。
This study aimed to establish a determination method of diester-alkaloids in Aconitum szechenyianum and Aconitum naviculare. The chromatographic method was carried out on the AichromrMC18 column using CH3OH- 0.05mol· L^-1 KH2PO4-CH3COOH- (CH3)2CHOH (67:173:4:4) as a mobile phase. The detection wavelength was set at 230nm, with the flow rate of 1.0 mL-min-1 and the column temperature of 35℃ The aconitine calibration curves were found to be linear in the range of 0.09632-0.48161xg (r=0.9998). The average recovery of aconitine was 96.3% with RSD=1.87%. This method is simple and reliable, providing a reference standard for the quality control of aconitum alkaloids.
