摘要
以米曲霉作为原始菌株,通过在纤维素培养基上驯化,经硫酸二乙酯(DES)、紫外线照射复合诱变,确定最适复合诱变条件为:用0.1ml15%DES处理后,紫外线照射用30W紫外灯,垂直距离20cm,照射时间为60s,得到一株高产纤维素酶的突变菌株Ⅱ5,其纤维素酶活力高达100.23U/100ml,为出发菌株的1.39倍。
Aspergillus oryzae was selected as the initial strain for breeding cellulase-producing mutants. A mutant strain, C3 was obtained through combinatorial mutagenization with UV and DES. The optimal mutation conditions were sequential treatment with 0.1 ml of 15% DES and then illumination with 30 W UV lamp for 60 seconds at a distance of 20 cm. The ceUulase activity of this high-yield strain reached 100.23 U/100 ml, which was 1.39 times of that of initial strain.
