摘要
目的:研究黑灵芝多糖对D-半乳糖诱导的衰老小鼠的抗衰老作用,并探讨其可能作用机制。方法:小鼠随机分为6组:空白对照组,D-半乳糖衰老模型组(100mg/kgbw·d),VC组(100mg/kgbw·d),黑灵芝多糖低、中、高剂量组(50、100、150mg/kgbw·d)。10周后,测定小鼠体重和肝脏指数,测定小鼠肝脏组织和脑组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、过氧化氢酶(CAT)活性及丙二醛(MDA)含量。结果:与模型组比较,黑灵芝多糖可拮抗小鼠体重的降低和肝脏的萎缩、增强肝脏组织和脑组织SOD、GSH-PX和CAT活性、减少MDA含量。结论:黑灵芝多糖具有延缓衰老的作用,其作用机制可能与抑制自由基,增加体内抗氧化酶的活性,减少过氧化脂质的生成,提高机体抗氧化能力有关。
Objective: To study the anti-ageing effect of polysacchadde from Ganoderma atrum (PSG-1) on aging mice induced by D-galactose and explore the underlying mechanisms. Methods: Healthy female mice were randomly divided into 6 groups: control group, aging model group, vitamin C (VC) (positive control) group (100 mg/kg · d), and low-, medium- and high-dose PSG-1 groups (50, 100, 150 mg/kg · d). After 10 weeks, the body weight and the liver index were examined and the activities of super-oxide dismutase (SOD), glutathione peroxidase (GSH-PX) and catalase (CAT) and the maleic dialdehyd (MDA) content were measured respectively in liver tissue and brain tissue of mice. Results: In comparison with the aging model group, the activities of SOD, GSH- PX and CAT in liver and brain tissue of the three PSG-1 groups all were increased and the MDA content was decreased and the loss of body weight and the liver atrophy markedly were counteracted. Conclusions: PSG-1 has antiaging effects on aging model mice and the mechanism may be related to its inhibitory effect on free radicas, the enhancement of activity of antioxidases and the reduction of lipid peroxidation.
