摘要
目的探讨蟾蜍灵对脂多糖(LPS)诱导的大鼠肾小球系膜细胞(GMC)增殖及白细胞介素6(IL-6)和转化生长因子β1(TGF-β1)表达的影响。方法体外培养大鼠肾小球系膜细胞,分为蟾蜍灵干预(A)组、LPS刺激(B)组和对照(C)组,应用台盼兰拒染法检测蟾蜍灵对GMC的细胞毒性作用,MTT观察LPS诱导的GMC增殖的变化,RT-PCR检测TGF-β1和IL-6 mRNA的相对表达。结果与B组相比,A组蟾蜍灵10-8mol/L和10-7mol/L明显抑制GMC增殖,A492nm值分别为[(0.818±0.058)和(0.574±0.182),P<0.01]。B组IL-6和TGF-β1 mRNA相对表达量较C组和A组明显减少(P<0.01)。结论蟾蜍灵明显抑制LPS诱导的大鼠系膜细胞增殖及其IL-6和TGF-β1的表达。
Objective To study the effects of bufalin on expressions of interleukin 6(IL-6) and transforming growth faetor-β1 (TGF-β1) in glomerular mesangial cells (GMCs) of rats. Methods GMC lines of rats were cultivated. GMCs in vitro were divided into 3 groups of bufalin intervention group(A) (bufalin 1 × 10^-8 mol/L), lipopolysaccharide(LPS) group(B) ( LPS 5 mg/L) and control group(C). Proliferation of GMCs was assessed by MTT assay and the mRNA of IL-6 and TGF-β1 was measured by RT-PCR. Results GMC proliferation induced by LPS was inhibited dose-dependently by 10^-8 mol/L and 10^-7 mol/L (A492nm values were 0. 818±4 0. 058 and 0. 574± 0. 182, respectively, P〈 0.01) in group A. The expressions of IL-6 and TGF-β1 mRNA were significantly less in group A than those in groups of C and B (P〈. 0.01). Conclusion Bufalin inhibits the GMCs proliferation and expression of IL-6 and TGF-β1.
