玉屏风多糖对大鼠肺纤维化的干预作用及部分机制研究 被引量:7

摘要 目的研究玉屏风多糖(YPF-P)对大鼠肺纤维化的干预作用。方法30只Wistar大鼠,随机分为假手术对照组、模型组、YPF-P 60 mg/kg组、YPF-P 120 mg/kg组、强的松组。除假手术对照组外其余各组大鼠气管内一次性滴注盐酸平阳霉素诱导肺纤维化模型,假手术对照组大鼠用生理盐水代替。次日各组分别给予YPF-P 60、120 mg/kg及强的松3.22 mg/kg灌胃给药,假手术对照组、模型对照组用生理盐水代替。给药后第28天处死全部动物。观察各组肺间质纤维化形成的病理变化;检测大鼠肺组织羟脯氨酸(Hyp)含量;血清放免法测定Ⅲ(Col-Ⅲ)、Ⅳ(Col-Ⅳ)胶原、层黏连蛋白(LN)、透明质酸(HA)的含量;RT-PCR法观察各组大鼠肺组织TGF-β1mRNA的表达。结果与模型组比较,YPF-P组病理变化较模型组减轻,YPF-P能显著降低大鼠肺组织Hyp和血清中Col-Ⅲ、Col-Ⅳ、LN、HA的含量,并能下调大鼠肺组织TGF-β1mRNA表达。结论YPF-P可以改善盐酸平阳霉素诱导的肺纤维化大鼠的一般情况,并通过抑制大鼠肺组织TGF-β1mRNA表达,以达到抗纤维化的作用。 Objective To study the effects and mechanism of Yu-Ping-Feng-Powder polysaccharide (YPF-P) on pulmonary fibrosis in rats. Methods 30 wistar rats were randomly divided into the sham operation group, the model group, Yu-Ping-Feng-Powder polysaccharidc 60 mg/kg-treated group, Yu-Ping-Feng-Powder polysaccharide 120 mg/kg-treated group and prednisone group. Rats in the model group, Yu-Ping-Feng-Powder polysaccharide 60 mg/ kg-treated group, Yu-Ping-Feng-Powder polysaccharide 120 mg/kg-treated group and prednisone group were injected with a single dose of BleomycinA5 by trachea, and rats in sham operation group with the same normal saline. After 24 h, 120,60 mg/kg was respectively given to rats daily by gastrogavage, while equal volume of normal saline was given to those in the sham operation group and model group, and an equal volume of prednisone (3.22 mg/ kg). On day 28, all rats were killed, the levels of Ⅲ-collagen, Ⅳ-collagen, laminin and hyaluronic acid in the serum were determined, the determination of hydroxyproline in lung homogenates were analyzed, and the lung was incised to make pathological sectons which were stained with HE and Masson. Expression of TGF-1 was detected by RT-PCR. Results Yu-Ping-Feng-Powder polysaccharide could significantly decrease the levels of Ⅲ-collagen, Ⅳ- collagen, laminin and hyaluronic acid in serum, concentration of hydroxyproline (Hyp) and expression of TGF-1 mRNA. Lung histology in the Yu-Ping-Feng-Powder polysaccharide treated group was also improved. Conclusion Yu-Ping-Feng-Powder polysaecharide can inhibit the effect on pulmonary fibrosis induced by Bleomycin A5 possib through decreasing the expression of TGF-β1 mRNA.
