皂角提取物对急性脑缺血再灌注损伤大鼠的保护作用 被引量:4

Protective effect of the extractive of Gleditsia sinensis Lam to the rats during acute cerebral ischemia reperfusion injury

摘要 目的观察皂角提取物对急性脑缺血再灌注损伤的保护作用。方法将SD雄性大鼠随机分为假手术组、脑缺血再灌注损伤模型组、复方丹参组及皂角提取物大、中、小剂量组(150、75、38 mg.kg-1.d-1)。用药组术前连续给药3 d,然后用线栓法造模,假手术组仅分离右侧颈总动脉、颈外动脉和颈内动脉,然后缝合伤口,其余各组大鼠分别于术后2 h拔线实现脑血流再灌注,24 h后观察神经功能评分,比较各组大鼠血清中丙二醛(MDA)和脑组织匀浆中NO以及髓过氧化物酶(MPO)的含量。结果皂角提取物能有效改善大鼠脑缺血再灌注后的神经功能,降低NO、MDA、MPO的含量。结论皂角提取物对急性脑缺血再灌注损伤有保护作用。 OBJECTIVE In order to observe the potential protective effect and possible mechanism of extractive of Gleditsia sinensis Lam (GSL) to the rats during focal cerebral ischemia - repeffusion(I/R) injury. METHODS 36 SD rats were randomly divided into 6 groups : sham - operation group, cerebral ischemia - reperfusion group, complex salvia Mihiorrhiza group ( 32 mg· kg^ - 1 · d^ - 1 ) , extractive of GSL groups with different doses ( 150,75,38 mg· kg ^-1 · d^-1 ). Transient middle cerebral artery (MCA) occlusion was induced as described by Longa. In sham - operation group,the skins of rats were only sewn up after exposing the common carotid artery (CCA) in the right side. After 2 hours of cerebral ischemia,the rats persisted for 24 hours of reperfusion,then, scores of neurological deficit were observed and NO, MDA, MPO levels were detected by chemical methods. RESULTS Extractive of GSL can improve scores of neurological deficit, and release the levels of NO, MDA, MPO. CONCLUSION Extractive of GSL perform a protective effect to the rats during focal cerebral ischemia - repeffusion(I/R) injury.
