摘要
对玉米蛋白粉叶黄素酶解辅助提取工艺进行了研究,采用响应面分析法分析了酶浓度、底物浓度和酶解时间对总叶黄素得率的影响,并建立了相应的预测模型。方差分析的结果表明,酶解时间的二次方项(P<0.025)和底物浓度的二次方项(P<0.005)对提取效率有显著影响,3个因素对提取效率也都有显著影响(P<0.005),并且酶浓度和底物浓度及酶浓度和酶解时间之间存在着交互作用(P<0.01)。考虑到提取效率、成本和可行性,利用约束复合形法对结果进行优化,得到的最佳工艺参数为:酶浓度7 682 U/g、底物浓度8.8%、酶解时间2.2 h。在最佳工艺参数下,总叶黄素的得率为64.65μg/g,与预测结果相符。与常规提取方法比较,叶黄素得率显著提高(P<0.05)。
Technology for enzyme treatment extraction of xanthophylls from corn gluten meal was investigated. Response surface methodology was used to analyze the effects of enzyme concentration, substrate concentration and hydrolysis time on total xanthophyll yield, and built the prediction model. The T - test and P - value indicated that the quadratic of hydrolysis time ( P 〈 0. 025 ) and the quadratic of substrate concentration ( P 〈 0.005 ), as well as the three variables were important linear variables affecting the total xanthophylls yield ( P 〈 0. 005 ) ; the interaction effect between enzyme concentration and substrate concentration, and the interaction effect between enzyme concentration and hydrolysis time were significant (P 〈 0.01 ). Considering efficiency, cost of materials, and experiment feasibility, the technique parameters were optimized by the constrained complex method. The optimal conditions obtained are as follows: enzyme concentration, substrate concentration and hydrolysis time are 7682 U/g, 8.8% and 2.2 h, respectively. The highest total xanthophylls yield is analyzed as 64.65 μg/g when corn gluten meal is hydrolyzed under the optimal conditions, which is consistent with the predicted value by RSM. Comparing with the traditional extraction method, the yield is increased significantly (P 〈 0. 05 ).
