摘要
目的研究星状神经节阻滞(SGB)对大脑缺血再灌注损伤的影响。方法雄性Wistar大鼠72只,体质量220~280g,随机分为3组,分别为正常组(n=8)、对照组(n=32)和实验组(n=32)。正常组实施假手术;对照组和实验组组均制作左大脑中动脉缺血(MCAO)再灌注损伤模型;实验组组在大鼠脑缺血再灌注后30min即行持续星状神经节阻滞。脑缺血后8h、16h、24h、32h灌注取材观察表达缺氧诱导因子1α(HIF-1α)细胞数及神经细胞凋亡数。结果对照组、实验组各时间点HIF-1α神经细胞凋亡蛋白的表达均明显高于正常组(P<0.05);实验组各时间点表达HIF-1α细胞数和神经细胞凋亡数明显低于对照组(P<0.05)。结论SGB能引起脑缺血再灌注损伤鼠HIF-1α及神经细胞凋亡蛋白的表达下调,对大鼠脑缺血再灌注损伤有保护作用。
Objective To investigate the effects of stellate ganglion block (SGB) on the expression of hypoxia inducible factor - 1 ( HIF - 1 α) and apeptosis following cerebral ischemia - reperfusion injury in rats. Methods Seventy - two male Wistar rats weighing 250 - 280 g were randomly divided into three groups : control group ( n = 32), sham operation group ( n = 8 ) and SGB group ( n = 32 ). HIF - 1α expression and apoptesis were investigated at 8 h, 16 h, 24h and 32 h pest - operation in middle cerebral artery occlusion (MCAO) rats in control group. The rats in sham operation group were exposed but not infracted. Left stellate ganglion was blocked 30 nfinutes after reperfusion in MCAO rats, then HIF - 1α expression and apoptosis at 8h , 16h , 24h and 32h post - operation were investigated in SGB group. Results The expression levels of HIF - 1α and apoptosis protein were markedly higher than those in sham operation group at different time points in both control group and SGB group ( P 〈 0.05 ). The number of HIF - 1 α expression cells and the number of apeptotic cells were significantly lower in SGB group compared with those in control group ( P 〈 0.05 ). Conclusion The study suggests SGB may depress the over - expression of HIF - 1 α and apoptosis induced by cerebral ischemia - reperfusion.
