安肠愈疡汤口服合加味生肌散灌肠对实验性溃疡性结肠炎大鼠的影响

Study on effect and mechanism of Anchang Yuyang Decoction with Jiawei Shengji Powder on ulcerative colitis in rats

目的:观察安肠愈疡汤口服合加味生肌散灌肠对实验性溃疡性结肠炎(UC)大鼠的疗效及机制探讨。方法:将90只SD大鼠随机分为6组[正常对照组、模型对照组、安肠愈疡汤组、加味生肌散组、安肠愈疡汤合加味生肌散组、柳氮磺吡啶(SASP)组]。除正常对照组外,其他各组均采用2,4,6-三硝酸苯磺酸复制UC模型,给药3周后,进行结肠大体形态和组织损伤评分,测定结肠组织内髓过氧化物酶(MPO)的活性以及白介素-1β(IL-1β)、白介素-8(IL-8)、肿瘤坏死因子(TNF-α)、白介素-10(IL-10)的含量。结果:各试验组大鼠结肠黏膜均有不同程度的炎症修复、溃疡愈合。结肠大体形态和组织学损伤评分及MPO活性均不同程度降低,结肠黏膜中IL-1β、IL-8、TNF-α含量也不同程度降低,而IL-10含量升高,与模型对照组相比有显著性差异(P<0.05或P<0.01),尤其以安肠愈疡汤合加味生肌散组最显著。结论:安肠愈疡汤合加味生肌散具有较好的抗炎和促进溃疡修复的作用,并能下调致炎细胞因子和上调抗炎细胞因子,从而使免疫功能恢复。

Objective：To study effect and mechanism of Anchang Yuyang Decoction with Jiawei Shengji Powder （ACYYDJWSJP） on ulcerative colitis （UC） in rats. Methods： Ninety SD rats were divided randomly into six groups： normal control group,pathologic model control group,ACYYD group, JWSJP group, ACYYD JWSJP group And Solfasalazine （SASP） treatment group. The model of ulcerative colitis were established by given 2,4,6-TNBS. After medication for 3 weeks, the parameters including colonic gross morphology, colon tissue injury scores, MPO activity, the content of IL-1β, IL-8, TNF-α and IL-10 were observed. Results： Inflammation relieving of colonic mucosa and ulcer healing were found in all treatment group rats at different degree. Colonic gross morphology, colon tissue injury scores,MPO activity and the level of IL-1β,IL-8, TNF-α decreased, while levels of IL-10 increased after therapy （P0.05 or P0.01）, and there was significant difference among groups,especially between ACYYD JWSJP treatment group and pathologic model control group. Conclusion： Research showed that ACYYD JWSJP had made a good effect. The mechanism might be down-regulating the expressions of pro-inflammatory cytokines （IL-1β,IL-8,TNF-α） and upregulating the expressions of anti-inflammatory cytokine （IL-10）, which can then adjust the immune system, inhibit the inflammation reaction and promote ulcer healing.
