摘要
为了更好地利用花药培养为杂交水稻遗传研究和育种服务,必须得到较大群体的二倍体花粉绿苗,通过改良培养基来提高花粉绿苗的加倍率是最主要的途径。为了评价改良培养基的效果,笔者引入一个新概念:二倍体得苗率(二倍体得苗率=移栽成活率×二倍体率=移栽成活的二倍体绿苗数/总试管苗数)。本试验在生根培养基中加入2.5mg/L多效唑和4mg/L秋水仙碱得到的二倍体得苗率显著高于加入4mg/L秋水仙碱的生根培养基和基本的生根培养基。结果表明,多效唑对秋水仙碱染色体加倍有增效作用。
In order to make better use of anther culture of hybrid rice for genetic research and breeding, a large group of diploid pollen green plants must be gotten. The best way is to enhance pollen seedlings doubling rate by improving culture media. To evaluate the contribution of improved culture media, we introduced a new concept- achievement rate of doubled haploid ( achievement rate of doubled haploid = tansplant survival frequency * Doubled haploid frequency = survival diploid green seedlings/total green seedlings). The result showed that the frequency of survival diploid green plants that had the rooting media with 2.5mg/L MET and 4mg/L colchicine was significantly increased than that of only 4mg/L colchicines in rooting culture medium or general rooting medium. This indicated that TEM had a promoting effect with colchicines on chromosome doubling.
