摘要
正常妊娠6~8周人绒毛滋养层细胞以Percol分离、纯化及体外培养为实验手段,观察马鞭草提取液对滋养层细胞功能的影响,进一步探讨了马鞭草抗早孕的细胞学作用机理。结果表明:25g/L、50g/L的马鞭草提取液能阻碍细胞生长,培养液中人绒毛膜促性腺激素(HCG)含量明显减少。琥珀酸脱氢酶(SDH)组织化学及HCG免疫组织化学染色,NYD-1000显微图像分析系统定量分析显示,细胞内SDH的活性及HCG分泌有明显抑制作用,其抑制率随加药剂量增加而增大。超微结构观察也显示细胞明显受损。一定浓度的马鞭草提取液能干扰细胞能量代谢,抑制HCG的合成和分泌。
To study the mechanism of abortifacient effect of extract of Verbena officinalis (VO), trophoblast cells from human placenta of 6 ̄8 weeks early pregnancy was isolated with Percoll method and cultured in vitro. The results showed that VO at 25 g/L, 50 g/L inhibited not only cell growth but also hCG secretion significantly. After effects of VO for 48 h, the trophoblast cells were stained with succinate dehydrogenase (SDH) histochemical and hCG immunohistochemical methods. These stained slides were quantified by NYD 1000 microimage analysis system. The average OD of SDH and hCG positive reactive intensity range of treated groups were significantly less than that of the control group ( P <0.01). SDH activity and hCG expression were inhibited by VO in dose dependent manner. Pathological changes including decreased microvilli, enlarged endoplsamic reticular cisternae, myelin like changed mitochondria and condensed chromatin arranged below nuclear membrane were also shown in the ultrastructures of trophoblast cells in treated group. Conclusively, these data in present study suggested that VO of certain concentrations directly damaged trophoblast cells and hindered the energy metablism of cells. These changes led to decreased hCG expression. It provided some theoretical evidence for the clinical abortifacient application of VO.
