摘要
采用ConA诱导的实验性小鼠肝损伤模型观察国产大剂量胸腺肽对小鼠肝脏的保护作用及对肝细胞凋亡的影响.结果表明:肝损害组与正常组ALT和TNF-α测定值有显著性差异,光镜检查肝损害组亦有严重的肝损伤;用大剂量胸腺肽腹腔注射一周后可明显减弱ConA对肝组织的损伤;对小鼠肝组织Fas抗原阳性细胞情况研究表明,肝损害组Fas抗原阳性细胞较为普遍,正常组无阳性细胞,胸腺肽组阳性细胞明显减弱.本实验结果提示大剂量胸腺肽对ConA诱导的肝损伤有保护作用;ConA可诱导肝细胞凋亡,大剂量胸腺肽对凋亡有抑制作用.
The effects of locally made thymic peptides on the Con A induced hepatic injuries and the apoptosis of hepatocytes were investigated in this study. The experimental results revealed that the plasma TNF-α and ALT levels were significantly higher in mice with hepatic injuries induced by Con A than those in normal mice; but when the mice were treated with large amount of thymic peptides, the hepatic injuries were remarkablly attenuated about one week after the treatment. Meanwhile, the Fas antigen positive cells were rather prominent in mice with hepatic injuries, and this kind of cells were significantly reduced after treatment with thymic peptides. These results suggested that the use of large amount of thymic peptides could protect theanimals from hepatic injuries induced by Con A, and have an inhibitory effect on the development of apoptosis of hepatocytes.
