不同产地巴戟天中糖类成分HPLC-ELSD指纹图谱研究 被引量:17

HPLC-ELSD Fingerprint of carbohydrate constituents in Morinda officinalis from different habitats

摘要 目的建立巴戟天中糖类化学成分HPLC指纹图谱的分析方法,研究不同产地的巴戟天药材的质量。方法采用HPLC法。色谱柱为Inertsil NH2(250mm×4.6mm,5μm);流动相:乙腈-水梯度洗脱;柱温:室温;体积流量:1.2mL/min,以蒸发光散射检测器(ELSD)检测,检测条件:漂移管温度100℃;N2气流速度2.2L/min;He在线脱气,气流速度20mL/min。使用计算机辅助相似度评价软件进行数据处理。对不同产地的巴戟天药材指纹图谱的相似度进行比较分析。结果除两批外,其余8批巴戟天与系统生成对照指纹图谱的相似度均在0.94以上,归纳出巴戟天13个共有峰。通过与对照品的保留时间比较,1、2号峰分别为D-果糖、蔗糖。不同产地巴戟天药材中糖类化合物化学组成相似,但相对比例有明显的差异。结论所用方法稳定、重现性好,可突出巴戟天药材的内在质量特征,为更好地控制巴戟天的内在质量提供了科学依据。 Objective To establish HPLC fingerprint analysis method of carbohydrate constituents in Morinda officinalis collected from different habitats. Methods HPLC Method was employed and a Columns Inertsil NH2 (250 mm×4.6 mm, 5 μm) with aeetonitrile-water as mobile phase by gradient elution was used, the flow rate was 1.2 mL/min, column temperature was room temperature. Detector was ELSD and the temperature of drift tube was 100 ℃. The flow rate of N2 was 2.2 L/min, Helium degasificatioh on-line with flow rate was 20 mL/min. The fingerprints evaluating software by computer-assisted for the similarity was used to process the experiment data and compare the similarity of HPLC fingerprint of M. officinalis from different habitats. Results Except for two batches, the fingerprint similarity value of other eight batches of herbs was above 0.94 compared with system generation spectrum and 13 common characteristic peaks were demonstrated. The first and second peaks were D-fructose and sucrose, respectively compared with the retention time of standard sample. The results showed that the carbohydrate constitutes of M. officinalis were similar among herbs from different habitats, but their relative ratios are significantly different. Conclusion The method in this experiment is stable and reproducible, which could distinguish the internal quality and provide the reference for quality control of M. officinalis.
