摘要
目的:建立反相离子对色谱法检测奶酪中三聚氰胺的含量。方法:采用乙酸铅与三氯乙酸作为蛋白沉淀剂对奶酪中的三聚氰胺进行提取,利用反相离子对色谱法,以乙腈和柠檬酸缓冲液为流动相,用10 mmol/L庚烷磺酸钠为离子对试剂,在240 nm的波长下对提取的三聚氰胺进行定性、定量的检测。结果:峰面积与三聚氰胺的含量在0.1-10μg/ml浓度范围内呈良好线性关系,相关系数r=0.9998,回收率在96.43%-101.71%之间,相对标准偏差均小于2%。最低检出限为0.1μg/ml。结论:该方法操作简便、灵敏、结果准确可靠,适合奶酪中三聚氰胺的检测。
Objective:Reversed -phase ion pair high performance liquid chromatographic method (RP -IP- HPLC) was established to determine melamine in cheese. Methods: Lead acetate and trichloroactic acid were used to extract melamine in cheese. Acetonitrile and citric acid buffer solution were used as mobile phase, 10 mmoL/L sodium heptanesulphonate as ion - paired reagent and detected at wavelength 240 nm. Results:In the range 0. 1 - 10 μg/ml of melamine, the peak area was linear with its concentration, with correlation coefficient(r) of 0. 9998, Recovery of melamine was 96.43% - 101.71%, relative standard deviation (RSD%) was less than 2%. The detection limits was 0. 1 μg/ml. Conclusion:The proposed method is simple, sensitive and reliable for the determination of melamine in cheese.
