胰岛素治疗对糖尿病大鼠膀胱M_2、M_3乙酰胆碱受体mRNA表达的影响 被引量:2

Effects of insulin treatment on mRNA expressions of M_2 and M_3 acetylcholine receptors in the urinary bladder wall of diabetic rats

摘要 目的检测链脲佐菌素诱导的DM大鼠模型膀胱乙酰胆碱受体mRNA表达水平的变化及胰岛素治疗的影响。方法实验大鼠分为正常对照组(NC组)、DM模型组(DM组)和胰岛素治疗组(DM+Ins组)。成模10周后取膀胱提取RNA,RT-PCR半定量分析3组大鼠膀胱乙酰胆碱M_2和M_3受体mRNA。结果与NC组相比,DM组和DM+Ins组M_2受体mRNA表达上调显著并以DM组最多,差异有统计学意义(P<0.01)。DM组和DM+Ins组M_3受体mRNA表达下调较NC组显著(P<0.01),尤以DM组明显(与DM+Ins组比较,P<0.05)。结论 DM大鼠膀胱M_2受体mRNA表达增加,M_3受体mRNA表达则减少,胰岛素治疗可能预防或减轻上述变化。 Objective To study the effects of insulin treatment on mRNA expressions of M2 and M3 acetylcholine receptors in the urinary bladder wall of diabetic rat. Methods Diabetic SD rat were induced by streptozotocin. The mRNA of M2 and M3 acetylcholine receptors in the rat bladder wall were measured with RTPCR at the end of 10th week. Results Compared with control group(1.13±0. 06), the M2 mRNA was highest in diabetic group(3. 86±0. 21) and higher in diabetic group with insulin treatment(1.58±0. 11) (P〈0. 01). And compared with control (0. 92±0. 04) the M3 mRNA was lowest in diabetic group(0. 26±0. 01) and lower in diabetic group with insulin treatment (0. 28 ± 0. 01 ) (P〈 0. 01 ). Conclusions Diabetic rat expresses the higher M2 mRNA and lower M3 rnRNA, and the expressions can be reduced by insulin treatment.
