内质网应激蛋白介导运动后骨骼肌钙稳态调节的研究 被引量:7

摘要 目的:研究内质网应激蛋白CRT对运动后细胞内钙稳态紊乱的调节作用。方法:使用改良的离心运动力竭模型为研究手段,SD大鼠被随机分成安静对照组(C)和单纯运动组(E),其中,单纯运动组再平均分为运动即刻组(E1)、运动后24h组(E2)、运动后48h组(E3)和运动后6d组(E4),每组8只。结果:运动后骨骼肌内质网,线粒体Ca2+浓度及Ca2+-ATP酶活性发生了明显的变化,离心运动后内质网E1组Ca2+浓度最高,与安静对照组(C)比较呈显著性差异(P<0.05),内质网Ca2+-ATP酶活性以E1组最高,与安静对照组(C)比较呈非常显著性差异(P<0.01);对线粒体来说,离心运动Ca2+浓度各时相以E2组Ca2+值最高,与安静对照组(C)比较呈非常显著性差异(P<0.01),E1及E3组值与安静对照组呈显著性差异(P<0.05),E3组值与E2组之间也呈显著性差异(P<0.05),同时E1组Ca2+-ATP酶值最高,与C组比较呈显著性差异(P<0.05),E3组值与C组比较也呈显著性差异(P<0.05);E2组比E1及E3组值都要低,并呈显著性差异(P<0.05),同时运动后E1、E2组骨骼肌中CRT的表达显著上升。结论:离心力竭运动后大鼠骨骼肌细胞内钙稳态发生失调,而运动后骨骼肌内质网应激蛋白的发生可能与运动后骨骼肌的钙紊乱有关,并对运动后细胞钙紊乱起到调节作用。 Objective: To investigate the endoplasmic reticulum stress protein of CRT on intracellular calcium homeostasis regulation disorder after exercise. Method:improved acute exhaustive eccentric exercise model would be applied, SD rats were randomly divided into control group quiet (C) and the simple exercise group (E), then the simple exercise group would be immediately divided into groups (E1) , 24-hour post-exercise group (E2), group 48 hours after exercise (E3) and six days post-exercise group (E4), n = 8. Results:after the eccentric exercise, the Ca^2+ concentration and Ca^2+-ATP activity in skeletal muscle endoplasrnic reticulum and mitochondria changed dramatically, after the eccentric exercise group, the Ca^2+ concentration of endoplasmic reticulum in group E1 had the highest value , with the quiet control group C relatively significant difference (P〈0.05), and the Ca^2+ -ATP activity had the highest value in E1 group, with quiet control group C much significant difference (P〈0.01), to mitochondria, after the eccentric exercise, the Ca^2+ concentration in the E2 group had the highest value , with the quiet control group C relatively significant difference (P〈0. 01), the value of E1 and E3 Group were significant differences (P〈0.05) with the quiet control group, the value of E3 group was also significantly different with the value of E2 (P〈0.05), at the same time, the Ca^2+ -ATP enzyme e in E1 group had the highest value with the C group significantly differences (P〈0.05 ), and E3 group compared with C group also showed significant difference (P〈0.05), the value of E2 had lower value with group E1 and he E3 group with significantly difference (P〈0. 05), at the same time , after exercise exhaust, the expression of skeletal muscle CRT in E1, E2 group increased significantly. Conclusion: the centrifugal force in rat skeletal muscle after exercise exhaust intracellular calcium homeostasis imbalance occurred, and skeletal muscle after exercise endoplasmic reticulum stress protein may be associated with the occurrence of calcium in skeletal muscle after exercise disorder related, and the movement disorder after the cells played the regulatory role of calcium.
