釉鞘蛋白在小鼠牙胚发育中的时空表达

摘要 目的:观察釉鞘蛋白(ameloblastin)在小鼠牙胚发育中表达的时空顺序及分布,探讨釉鞘蛋白在釉质发育矿化中的作用。方法:采用原位杂交的实验方法,观察釉鞘蛋白在牙胚发育的增殖分化期、分泌初期及分泌期的表达。结果:釉鞘蛋白mRNA在成釉细胞增殖分化期表达为阴性,分泌初期细胞内及新生釉基质中开始出现弱阳性表达;至分泌期,细胞内及新生釉基质中均呈强阳性表达。成熟釉质中无釉鞘蛋白mRNA的阳性表达。结论:本实验从分子水平上进一步明确了釉鞘蛋白的时空表达,并推测釉鞘蛋白可能介导了釉质的发育矿化,它与牙齿发育、釉基质矿化反应等过程密切相关。 Objective:To study the expression of ameloblastin in different development stages of Mus musculus tooth germ aud to disscuse the function of ameloblastin in the development of enamel. Methods:We performed in situ hybridization after preparing specimens of different stages of the incisor developing tooth germ in Mus musculus. Results:Ameloblastin mRNA was negative under microscope in the ameloblast proliferate differentiation period, it showed weakly positive expression in intra-cellular and new-birth enamel matrix in secrete initial stage while it showed strongly positive expression in intra-cellular and new-birth enamel matrix when it came to secretion phase. There was no positive expression of ameloblastin mRNA in mature stage. Conclusion : In this study, we further clarified the spatio-temporal expression of ameloblastin from the molecular level. The results suggest that ameloblastin may mediate the development of enamel matrices and mineralization process.
