摘要
目的建立大鼠非酒精性脂肪肝动物模型,为深入探讨非酒精性脂肪肝的发病机制及药物治疗提供实验参考。方法取Wistar大鼠60只,随机分为对照组15只(常规饮食喂养),模型组45只(高脂、高胆固醇饮食组)喂养,12周后处死动物取出肝脏,经H-E染色、组织化学与免疫组织化学染色观察造模情况。结果模型组大鼠肝细胞脂肪变性明显,eNOS免疫组化显示肝中央静脉和门管区内皮细胞呈强阳性。结论高脂、高胆固醇饮食是诱导非酒精性脂肪肝形成的重要诱因,降低其摄入是有效预防该疾病的重要措施,此外eNOS在病变早期起到减轻肝损伤的作用。
Objective To establish the model of rat nonalcoholic fatty liver(NAFL)and observed the oi'iginating mechanism and drug therapy of this disease. Methods 60 Male SD rats were randomly divided into two groups:control group( n= 15 ),fed chow diet;and model group( n=45 ), these rats were fed high fat diet. After 12 weeks, all rats were sacrificed, and their livers were taken for histology observation. The results were detected by basic histology, cell chemistry and immunohistochemistry. Results High fat and cholesterol diet can significantly increase nonalcoholic fatty liver,and increas the expression of eNOS. Conclusion High fat and cholesterol diet were an important factor inducing nonalcoholic fatty liver; eNOS may play an important role in preventing the damages of liver cell in NAFL.
