超微白参对脾虚小鼠小肠上皮细胞蛋白质组的比较研究

Difierential Study of Dry Radix Ginseng of the Supermicro-Medicinal Slices on Proteomics ot Small Intestinal Epithelia of the Splenasthenic Syndrome Mice

目的:探讨白参超微饮片与其传统饮片的量效关系,了解白参对脾虚小鼠小肠上皮细胞蛋白质组调节的作用靶点。方法:选用昆明种小鼠120只,雄雌各半,利血平法复制动物模型,随机分为正常、脾虚模型、白参传统汤剂组、超微白参等剂量组、1/2剂量组、1/4剂量组,造模成功后分别灌服相应供试液,用双向凝胶电泳技术分离蛋白质,改进的考染法染色,以PDQuest软件分析图谱,用MALDI-TOF-MS进行质谱分析。结果:在回调点数、回调率、回调到正常点数、回调正常率几方面,超微白参1/2剂量组、超微白参等剂量组与白参传统汤剂组相近,超微白参1/4剂量组则低于白参传统汤剂组,但其差异尚未显示出统计学意义。结论:超微白参1/2剂量具有与白参传统汤剂相同的健脾疗效;白参健脾是通过多途径的、多靶点来实现的,主要针对小肠上皮细胞第3、4、10、20、23、27、28、31号蛋白点的调节达到健脾作用。

Objeetive： To approach the dose - effectiveness relationship between dry Radix Ginseng of the Supermicro - medicinal slices and traditional medicinal slices. To approach the functionary targets of dry Radix Ginseng of Chinese traditional medicine on proteomics of small intestinal epithelia of the Splenasthenic Syndrome mice. Methods;The 120 Kunming Mice,male and female half and half, were divided randomly into 6 groups：normal group,medel group,dry Radix Ginseng decoction group,micropowder dry Radix Ginseng equal dose group,micropowder dry Radix Ginseng 1/2 dose group and micropowder dry Radix Ginseng 1/4 dose group. Which were duplicated Splenasthenic Syndrome animal model by reserpine and then were given corresponding laboratorial medicine. Two - dimensional polyaerylamide gel electrophoresis technology was applied to separate proteins of small intestinal epithelia of mice of every group. The proteins spots were visualized with an improved technique of Coomassie brilliant blue staining;the pictures of proteins of every group were an- alyzed with the PDQuest software;The mass spectrometry analysis was used with MALDI - TOF - MS. Results：Their curative effect of micropowder dry Radix Ginseng 1/2 dose group, micropewder dry Radix Ginseng equal dose group and traditional dry Radix Ginseng decoction group is similar in the numbers of spots of regulated, hack. the regulated - back percent, the number of spots of regulated - back to normal,the percent of regulated - back to normal. Its curative effect of micropowder dry Radix Ginseng 1/4 dose group is under dry Radix Ginseng decoction group in these aspects and the statisti- cal significance of their difference was not showed. Conclusion： Micropowder dry Radix Ginseng 1/2 dose has same invigorating splenic effect of the accustomed remedial method of dry Radix Ginseng. Radix Ginseng come true its invigorating splenic function by its many routes and many targets ,especially by regulating number 3,4,10,20,23,27,28,31 proteins spots of small intestinal epithelia.
