糖心康对糖尿病大鼠心肌组织基因Pank1mRNA表达的影响

摘要 目的:比较糖尿病性心肌病大鼠和中药复方糖心康治疗组大鼠心肌组织中基因表达的差异,并用Evegreen实时荧光RT-PCR技术定量检测泛酸激酶1(Pank1mRNA)在各组大鼠心肌组织的表达水平。方法:(1)差异表达基因的筛选;(2)荧光定量RT-PCR的检测。结果:该文仅对筛选出差异表达基因谱中的Pank1mRNA进行报告。糖尿病性心肌病模型组心肌组织基因Pank1mRNA表达下调,中药复方糖心康治疗组心肌组织基因Pank1mRNA表达上调。结论:Pank1mRNA在糖尿病状态下心肌组织也有表达,并且表达下调,而中药复方糖心康可使其上调,这也是中药复方糖心康对糖尿病性心肌组织损伤具有保护作用的机制之一。 Objective: To study the difference of myocardial tissues gene - expression between diabetic rats and rats therapied by herb prescription. To measure Pankl mRNA - expression of myocardial tissues with eve green real time RT - PCR. Methods : ( 1 ) To choice the different expression gene; (2) To measure the production with eve green real time RT- PCR. Results: Get different expression Gene-Pank1. The expression of diabetic eardiomyopathy diabetes mellitus gene - Pankl is decreased, while the expression of rats myocardial gene - Pankl mRNA therapied by Tangxinkang is increased. Conclusion: The paper only reported one gene - Pankl mRNA of rat myocardial gene expression profiles. Myocardial tissues gene - Pankl mRNA is also expressed when suffering from diabetic and the number of expression is less than normal. While TangxinKang can make the number of this gene expression increase. This is one of protection mecha- nisms of diabetic myocardial tissues damage therapied by herb prescription.
