四妙勇安汤对实验性腹主动脉瘤生长的抑制作用及机理 被引量:6

摘要 目的:探讨四妙勇安汤对实验性腹主动脉瘤(Abdominal Aortic Aneurysm,AAA)生长的影响及机制。方法:建立大鼠AAA模型,随机分成四妙勇安汤组和盐水对照组,造模后2周、4周,应用弹力纤维染色、CD68免疫组织化学染色观察动脉壁弹力纤维的损伤、巨噬细胞的浸润,用RT-PCR和Western蛋白质印迹方法观察单核细胞趋化蛋白-1(monocyte chemotacticprotein-1,MCP-1)及基质金属蛋白酶-2(matrix metalloproteinases-2,MMP-2)的基因表达。结果:与盐水对照组相比,造模后2周时,四妙勇安汤组大鼠腹主动脉的扩张、动脉壁弹力纤维的损伤、CD68阳性表达、MCP-1和MMP-2的基因表达程度均明显降低(P<0.01)。结论:四妙勇安汤能通过抑制动脉壁炎性细胞的浸润和相关基因的表达,对实验性腹主动脉瘤的生长产生明显的抑制作用。 Objective :To investigate the inhibition and its mechanism of Simiao Yong'an Decoction (SMYAD) on experimental abdominal aortic aneurysm. Methods : To establish rat AAA models, all rats were randomly divided into SMYAD group and blank control group. At 2 weeks and 4 weeks after operation, elastic fiber staining and CD68 immunohistochemical staining were used to observe the injury of elastic fibers and the infiltration of maerophages in the arterial wall, RT - PCR and Western blot were used to observe the gene expressions monocyte chemoattraetant protein - 1 ( MCP - 1 ) and matrix metalloproteinases - 2 ( MMP - 2), respectively. Results : Compared with the control group, the expansion of the abdominal aorta, the injury of elastic fibers in the arterial wall, CD68, MCP - 1 and MMP - 2 gene expression on SMYAD group were all significantly reduced at 2 weeks after operation ( P 〈 0.01 ). Conclusion : SMYAD has the obvious inhibiting effects on the growth of experimental abdominal aortic aneurysm by inhibiting the inflammatory cell infiltration and related gene expression in the arterial wall.
