摘要
目的:探讨外周血单个核细胞(PBMCs)γ干扰素(IFN-γ)和白细胞介素-10(IL-10)mRNA水平在肝细胞癌(HCC)发病中的作用。方法:分离20例HCC患者和20例健康体检者外周血PBMCs,提取总RNA,利用半定量RT-PCR方法逆转录并扩增IFN-γ和IL-10,通过凝胶图像扫描系统对PCR产物的电泳条带进行密度扫描。结果:HCC患者PBMCs中IL-10mRNA水平明显高于健康体检者(P<0.01),而IFN-γmRNA水平明显低于健康体检者(P<0.01)。结论:肝癌患者外周血PBMCs存在Th1/Th2失衡。
Objective: To analyze the expression levels of interferon gamma (IFN-γ) and interleukin 10 (IL-10) mRNA in peripheral blood mononuclear cells (PBMCs) in the pathogenesis of hepatocellular carcinoma (HCC). Methods: PBMCs were isolated from 20 patients with HCC and the total RNA was extracted. The expression levels of IFN-γ and IL-10 mRNA in PBMCs were amplified by using semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR). The density of electrophoresis band in PCR products was scanned by gel scanning system. The results were compared with that from normal healthy volunteers. Results: RT-PCR analysis showed that the expression levels of IL-10 mRNA were significantly increased in patients with HCC compared with that in healthy individuals (P〈0.01). Whereas IFN-γ mRNA levels were significantly decreased compared with that in heahhy individuals (P〈0.01). Conclusion: Th1/ Th2 is imbalance in PBMCs with HCC.
