武汉地区水痘带状疱疹病毒基因型分析 被引量:9

Genotype analysis of varicella-zoster virus in Wuhan

摘要 目的探讨武汉地区水痘带状疱疹病毒流行的主要基因型。方法收集96例水痘、带状疱疹患者皮损标本。采用煮沸、冻融方法提取标本中的DNA。多重PCR扩增水痘带状疱疹病毒ORF22、38、54,并测序。用ClustalW软件比对和Primerpremier 5软件酶切位点分析核酸序列。结果多重PCR检测水痘、带状疱疹患者皮损标本阳性率为81.3%(78/96)。与Dumas序列进行对比,78份阳性标本ORF22在37902、38055、38081、38177存在点突变,符合基因型J型特点。ORF38和ORF54分别存在PstⅠ和BgⅡ酶切位点。结论武汉地区水痘带状疱疹病毒流行的主要基因型为J型、ORF38(PstⅠ^+)和ORF54(BgⅡ^+)。 Objective To determine dominant genotypes of varicella-zoster virus (VZV) in Wuhan. Methods Specimens were collected from lesions (vesicles or pustules) of 96 patients clinically diagnosed with varicella or herpes zoster, DNA was extracted from these specimens by boiling, freezing and thawing procedure. Then, multiplex PCR was performed to amplify the open reading frames (ORF) 22, 38, 54 of VZV followed by sequence analysis. Finally, the seqences were piled up with ClustalW programme and restriction sites were assessed with Primer premier 5 software. Results In total, 78 (81.3%) specimens were positive for multiplex PCR. The comparison with Dumas sequence revealed that all the positive specimens carried SNPs at position 37902, 38055, 38081 and 38177 in ORF22, which were consistent with J genotype. A recognition site was observed for PstⅠ and BgⅡ enzymes in ORF38 and ORF54, respectively. Conclusion In Wuhan, the major genotype of VZV is J with a PstI site in ORF38 and a BgⅡ site in ORF54.
