人脑胶质瘤组织p57^(kip2)和p16蛋白表达及其临床意义的研究

Expression and clinical significance of p57^(kip2) and p16 in human glioma

摘要 目的:探讨p57kip2蛋白、p16蛋白在人脑胶质瘤组织中的表达及其临床意义。方法:采用免疫组织化学SP法检测p57kip2、p16在55例人脑胶质瘤和10例正常脑组织中的表达。结果:在55例胶质瘤组织中,p57kip2蛋白阳性率为38.2%(21/55),显著低于正常脑组织80.0%(8/10),P=0.014。p57kip2蛋白表达在低度恶性(Ⅰ~Ⅱ级)组显著高于高度恶性(Ⅲ~Ⅳ级)组,P=0.029;生存时间≥2年组显著高于<2年组,P=0.032。脑胶质瘤组织中p16蛋白表达阳性率为50.9%(28/55),与正常脑组织相比差异无统计学意义,P=0.051。p16蛋白表达在高度恶性(Ⅲ~Ⅳ级)组显著高于低度恶性(Ⅰ~Ⅱ级)组(P=0.022),生存时间<2年组显著低于≥2年组,P=0.040。p57kip2蛋白的表达与p16蛋白的表达存在正相关,rs=0.388,P=0.003。结论:p57kip2蛋白和p16蛋白在脑胶质瘤中存在异常表达,p57kip2蛋白和p16蛋白的表达程度与脑胶质瘤的分化程度和预后相关。 OBJECTIVE:To explore the expressions of p57^kip2 and p16 in human gliomas and their clinical significance. METHODS: p57^kip2 and p16 were detected by SP immunohistochemical technique in 55 human glioma and 10 normal brain tissues. RESULTS:The expression rate of p57^kip2 protein in human glioma was 38.2 %(21/55), and significantly lower than that in normal brain tissues (80.0% ,8/10, P=0. 014). The p57^kip2 expression in low malignant group ( Ⅰ - Ⅱ grade) was significantly higher than that in high malignant group ( Ⅲ -Ⅳ grade, P=0. 029) and in the survival time ≥2 years group was significantly higher than that in the 〈2 years group (P=0. 032). The expression rate of p16 protein in human glioma was 50. 9 % (28/55), and was not significantly differrent from that in the normal brain tissue (P=0. 051). The p16 expression in the high malignant group( Ⅲ-Ⅳ grade) was significantly lower than that in the low malignant group ( Ⅰ - Ⅱ grade, P=0. 022) and in the survival time〈2 years group was significantly lower than that in the ≥2 years group (P=0. 040). There was a positive correlation between the expressions of p57^kip2 and p16 (rs=0. 388 ,P=0. 003). CONCLUSION: The expressions of p57^kip2 protein and p16 protein are deregulated in glioma, and their degrees are correlated with the glioma differentiation and prognosis.
